Primary antibodies specifically bind to the protein or biomolecule of interest and are directly conjugated to a dye or an enzyme for detection or detected with a secondary antibody. Splenocytes from the host are typically fused in vitro with cultured myeloma cells to produce immortal hybridoma cells with B cell characteristics. This allows for some hybridomas to continue to grow, synthesize, and secrete antibodies which specifically target the immunizing molecule. The hybridoma cells are subcloned through limiting dilution to identify a single clone which generates a monoclonal antibody to a single epitope on the immunogen. Monoclonal antibodies provide a stable, long-term supply of lot-to-lot consistent antibodies for a wide variety of applications including flow cytometry, ELISA, western blot, immunohistochemistry, and immunoprecipitation.
