What are primary antibodies?

Primary antibodies recognize and bind target antigens such as cell surface markers, signaling molecules, and post-translationally modified proteins with high specificity and affinity. This makes them useful for detecting and isolating various biomolecules with techniques including western blot, ELISA, immunoprecipitation, and flow cytometry.

Depending on how they are produced, primary antibodies are classified as either polyclonal or monoclonal. The main difference is that polyclonal antibodies recognize multiple epitopes, while monoclonal antibodies recognize just a single epitope. Both polyclonals and monoclonals can be labeled to allow for direct detection or can alternatively be paired with a suitable secondary antibody for indirect detection.

Primary antibodies are distinct from secondary antibodies, which recognize antibodies from a particular host species and of a specific isotype. Secondary antibodies are generally polyclonal and are named according to the host species and target species. For example, goat anti-mouse IgG1 secondary antibodies are raised in a goat and recognize mouse antibodies of the IgG1 isotype.

    Monoclonal antibodies are produced using hybridoma technology or recombinant methods. Their advantages include high specificity, consistent batch-to-batch performance, and stable long-term supply.

    Conjugated antibodies are widely used for multiplexed protein detection with techniques such as ELISA, immunohistochemistry (IHC) and flow cytometry. Available labels include enzymes, fluorescent dyes, and biotin.

  • Epitope Tag Antibodies
    Epitope tags are added to the N- or C- termini of proteins to facilitate their detection or isolation. They include short peptide sequences and larger protein molecules and are especially useful for studying targets for which specific antibodies are not available.
