Host Species Selection
For any custom polyclonal antibody production project, the choice of host species plays a pivotal role in achieving optimal results. At SouthernBiotech, we offer a diverse range of host species, each with unique characteristics and advantages tailored to your specific project requirements.
- Rabbit
Often used for generating antibodies against small and specific peptides or proteins. For instance, in research focused on studying human proteins, rabbits are preferred due to their ability to recognize subtle antigenic differences that might be missed in larger animals.
- Goat
Ideal for large-scale production of antibodies for use in diagnostic assays or therapeutic applications. Their ability to produce a large volume of antibodies makes them suitable for commercial antibody production or extended research studies.
- Sheep
Can be used in similar contexts to goats, especially for large-scale antibody production. An example is the production of antivenoms, where large quantities of antibodies against snake or spider venoms are required.
- Donkey
Sometimes used when a researcher needs large volumes of antibodies and other large animals like goats and sheep have already been used for immunization with different antigens. This helps to reduce the risk of cross-reactivity in experiments where multiple antibodies are used.
- Mice
Often used for generating polyclonal antibodies from serum when creating small-scale reagents for research or from ascites for customers requiring larger volumes for further manufacturing. Their use is particularly prevalent in basic scientific research, where the antibodies are used in techniques like Western blotting, immunoprecipitation, and immunohistochemistry.
- Swine
Sometimes chosen for antibody production against large proteins or complex antigens that might not be as immunogenic in other species.
The choice of host species is a critical decision influencing the success of antibody production. SouthernBiotech's expertise ensures that the selection aligns with your project goals, optimizing the antibody response, and overall project outcomes.
Polyclonal Antibody Generation
Antisera and ascites production is available on a large or small scale basis utilizing our standard protocols or a client supplied immunization procedure and schedule. Small pre-immunization serum samples are typically provided for screening. Upon completion of the protocol, the project may be terminated following your approval or continued on a monthly basis.
Antibody Purification
Antisera and ascites may be shipped separately, pooled following the final collections, or pooled and purified utilizing a selection of the antibody purification methods below:
- Affinity purification
- Protein G purification
- Protein A purification
- Cross-adsorption
- Ammonium sulfate precipitation
- Caprylic acid precipitation
- Size exclusion / Gel filtration chromatography
- Endotoxin removal
Antibody Conjugation
SouthernBiotech offers a variety of antibody conjugation options, to support your different assay needs, such as horseradish peroxidase (HRP), alkaline phosphatase (AP), biotin, β-galactosidase and fluorochromes including Alexa Fluor® dyes, FITC, PE, and PE/CY7.
Antibody Testing and Characterization
We can characterize your antibody using the following:
- Serum titer evaluation
- ELISA testing for specific activity
- Endotoxin testing by LAL assay
- HPLC
- Western blot
- Flow cytometry
- Immunohistochemistry
Antibody Production Protocols
We can use our standard protocols or customize timing to meet your specific project needs.
Time | Steps |
---|---|
Project Start | Exclusive use of animals, housing, maintenance, pre-immune bleed, and six (6) injections. |
Day 30 | First Collection |
Day 60 | Second Collection |
Day 90 | Third Collection |
Project Termination | All projects will be terminated ten (10) days after the last scheduled shipment unless we receive written instructions by Day 100 to continue maintaining the animals for a specified period at our per diem rate per animal. |
Project Extension | Upon your written instruction, we will extend your project at our per diem rate per animal. |
Note - We recommend that multiple animals are used in order to maximize the potential for a robust immune response. |
